Age- and Sex-Specific Plasticity in Dopamine Transporter Function Revealed by Food Restriction and Exercise in a Rat Activity-Based Anorexia Paradigm11/01/2019Eating disorders such as anorexia typically emerge during adolescence, are characterized by engagement in compulsive and detrimental behaviors, and are often comorbid with neuropsychiatric disorders and drug abuse. No effective treatments exist. Moreover, anorexia lacks adolescent animal models, contributing to a poor understanding of underlying age-specific neurophysiological disruptions. To evaluate the contribution of dopaminergic signaling to the emergence of anorexia-related behaviors during the vulnerable adolescent period, we applied an established adult activity-based anorexia (ABA) paradigm (food restriction plus unlimited exercise access for 4 to 5 days) to adult and adolescent rats of both sexes. At the end of the paradigm, measures of plasma volume, blood hormone levels, dopamine transporter (DAT) expression and function, acute cocaine-induced locomotion, and brain water weight were taken. Adolescents were dramatically more affected by the ABA paradigm than adults in all measures. In vivo chronoamperometry and cocaine locomotor responses revealed sex-specific changes in adolescent DAT function after ABA that were independent of DAT expression differences. Hematocrit, insulin, ghrelin, and corticosterone levels did not resemble shifts typically observed in patients with anorexia, though decreases in leptin levels aligned with human reports. These findings are the first to suggest that food restriction in conjunction with excessive exercise sex-dependently and age-specifically modulate DAT functional plasticity during adolescence. The adolescent vulnerability to this relatively short manipulation, combined with blood measures, evidence need for an optimized age-appropriate ABA paradigm with greater face and predictive validity for the study of the pathophysiology and treatment of anorexia. SIGNIFICANCE STATEMENT Adolescent rats exhibit a distinctive, sex-specific plasticity in dopamine transporter function and cocaine response after food restriction and exercise access; this plasticity is both absent in adults and not attributable to changes in dopamine transporter expression levels. These novel findings may help explain sex differences in vulnerability to eating disorders and drug abuse during adolescence. |
Analysis of electrical stimulation and voluntary muscle contraction on skeletal muscle oxygen uptake and mitochondrial recovery using near-infrared spectroscopy05/2023Purpose: This investigation was to compare differences in skeletal muscle oxygen consumption ([Formula: see text]) and mitochondrial recovery between voluntary (VOL) and electrically stimulated (ES) plantarflexion contractions. Methods: Twelve men and women (26 ± 4.0 years; 171.8 ± 5.1 cm; 74.0 ± 13.7 kg) were seated in a chair with their right knee fully extended and right foot secured to a force transducer. ES electrodes and a near-infrared spectroscopy device were placed on the gastrocnemius. Participants performed ES plantarflexion contractions across a range of stimulation intensities at frequencies of 1 and 2 Hz and similar VOL contractions. Cuff occlusion occurred immediately following each series of contractions to measure [Formula: see text]. A standardized mitochondrial function assessment protocol was also performed to calculate K-constants between work-matched ES and VOL contractions. Results: For mitochondrial assessments, there were no significant differences between ES and VOL rate constants (2.03 ± 0.98 vs. 1.25 ± 1.35 min-1, p = 0.266). ES resulted in a significantly greater workrate-[Formula: see text] slope at 1 Hz (0.007 ± 0.007 vs. 0.001 ± 0.002% [Formula: see text]/s/N, p = 0.014) and 2 Hz (0.010 ± 0.010 vs. 0.001 ± 0.001% [Formula: see text]/s/N, p = 0.012), as well as a significantly greater workrate-[Formula: see text] Y-intercept at 2 Hz (1.603 ± 1.513 vs. 0.556 ± 0.564% [Formula: see text]/s, p = 0.035) but not 1 Hz (0.579 ± 0.448 vs. 0.442 ± 0.357% mV̇O2/s, p = 0.535) when compared to VOL. Conclusion: ES results in a significantly greater [Formula: see text] at similar work rates compared to VOL, however, the mitochondrial recovery rate constants were similar. The greater mVO2 with ES may partially contribute to the increased rate of fatigue during ES exercise in individuals with muscle paralysis. |
Analysis of material motion characteristics of vertical nonlinear synchronous vibration dryer12/2019The nonlinear characteristics of a vertical dryer prototype under the synchronous excitation of vibration motors were analyzed. The vibration synchronization conditions of the two excitation motors of the vertical dryer were pursued. The analysis of motion characteristics of the material granules on the vertical dryer was conducted. The relationship between the structure parameters of the vertical dryer powered by two excitation motors and the motion characteristics of the material granules were discussed. Experimental results and discussion were used to verify the correctness of the theoretical analysis. This research demonstrated that the displacement of the material granules in the moving direction increased as the installment angle of the two excitation motors increased. This can improve the production efficiency; however, too large an installment angle destabilized the response of the dryer, no longer satisfying the design and use requirements. The research methods and conclusions presented in this paper can provide theoretical support and experimental basis for future development of a vibration dryer powered by two excitation motors. |
Analysis of Movement Entropy during Community Dance Programs for People with Parkinson's Disease and Older Adults: A Cohort Study01/07/2022Dance therapy can improve motor skills, balance, posture, and gait in people diagnosed with Parkinson's disease (PD) and healthy older adults (OA). It is not clear how specific movement patterns during dance promote these benefits. The purpose of this cohort study was to identify differences and complexity in dance movement patterns among different dance styles for PD and OA participants in community dance programs using approximate entropy (ApEn) analysis. The hypothesis was that PD participants will show greater ApEn during dance than OA participants and that the unique dance style of tango with more pronounced foot technique and sharp direction changes will show greater ApEn than smoother dance types such as foxtrot and waltz characterized by gradual changes in direction and gliding movement with rise and fall. Individuals participated in one-hour community dance classes. Movement data were captured using porTable 3D motion capture sensors attached to the arms, torso and legs. Classes were also video recorded to assist in analyzing the dance steps. Movement patterns were captured and ApEn was calculated to quantify the complexity of movements. Participants with PD had greater ApEn in right knee flexion during dance movements than left knee flexion (p = 0.02), greater ApEn of right than left hip flexion (p = 0.05), and greater left hip rotation than right (p = 0.03). There was no significant difference in ApEn of body movements (p > 0.4) or mean body movements (p > 0.3) at any body-segment in OA. ApEn analysis is valuable for quantifying the degree of control and predictability of dance movements and could be used as another tool to assess the movement control of dancers and aid in the development of dance therapies. |
Antisense Oligonucleotide Reverses Leukodystrophy in Canavan Disease Mice03/2020Marked elevation in the brain concentration of N-acetyl-L-aspartate (NAA) is a characteristic feature of Canavan disease, a vacuolar leukodystrophy resulting from deficiency of the oligodendroglial NAA-cleaving enzyme aspartoacylase. We now demonstrate that inhibiting NAA synthesis by intracisternal administration of a locked nucleic acid antisense oligonucleotide to young-adult aspartoacylase-deficient mice reverses their pre-existing ataxia and diminishes cerebellar and thalamic vacuolation and Purkinje cell dendritic atrophy. Anxiety and stress over COVID-19 pandemic associated with increased eating11/25/2021Objective Stressful experiences can dramatically affect eating. The relatively sudden, global emergence of the COVID-19 pandemic served as a massive stressor to virtually all people, regardless of infection status. This study hypothesized that actual and perceived stressors from the onset of the COVID-19 pandemic, in the categories of recurring disruptions, environmental threat, and social isolation would be positively associated with increased self-reported eating in the United States. Methods Over 1100 English-fluent adults (52.8% women) living in the United States were recruited for a cross-sectional online survey about eating, COVID-19 consequences, and stress experiences. Linear regressions examined associations between perceived stress on five eating measures, and individual differences in personal/work situations, perceptions, and adverse experiences during the pandemic. Results Anxiety, worry, and stress over, rather than direct consequences of, COVID-19 were most consistently associated with self-reported increased eating. Largely, these fell into the stressor categories of environmental threat and social isolation, not recurring disruptions. Body mass index and current self-reported eating pathology symptoms were also consistently associated with these outcomes. Conclusions These correlational findings suggest specific stressors have pronounced influences on eating behavior of US adults. Remotely deliverable stress mitigation strategies should be explored to attenuate increased eating. |
